Reactome-Recon connection

The connection was chosen from the Reactome-Recon lists: proteins of the Recon2 metabolic reconstruction that take part in Reactome signalling processes. nNOS (NOS1) is in the lists through the Reactome process ATP2B4 binds to NOS1, inhibiting it (R-HSA-5617178).

Description

Neuronal nitric oxide synthase (nNOS, NOS1) makes nitric oxide and citrulline from arginine, NADPH and O2 and is active only with Ca2+-bound calmodulin, so its output follows the Ca2+ signal of the cell. CaMKII phosphorylates nNOS and lowers its activity, a feedback that limits nitric oxide production in neurons; protein phosphatases 1 and 2A remove the phosphate. The plasma membrane Ca2+ pump PMCA4b (ATP2B4) binds nNOS and inhibits it; the inhibition needs both the binding and the Ca2+ export, which lowers Ca2+ next to nNOS.
